  报告人简介:

  Dr. YU obtained his bachelor degree from department of virology, Wuhan University in 2002 and Ph. D degree from department of biochemistry, Case Western Reserve University in 2007. After that, he carried out postdoctoral research in Case Western Reserve University and Cold Spring Harbor Laboratory from 2007 to 2010 and 2010 to 2016, respectively. From October, 2016 until now, he worked in Institute of Biophysics, Chinese Academy of Sciences.

  He has obtained several awards such as Travel Award from Mobile Genetic Elements, Best Poster Award at CSH-Asia Conference: RNA Biology and American Heart Association Postdoctoral Fellowship Award. He has published several papers in famous scientific journals such as Science, Cell and RNA as first author with more than 500 citations.

  Dr. YU Yang's research is focused on the study of biological functions and mechanisms of non coding nucleic acids (such as piRNA, miRNA, lincRNA).

  Protein-RNA interaction networks are essential to understand gene expression controls. Identifying the binding sites of RNA-binding proteins (RBPs) by CLIP (UV-crosslinking and immunoprecipitation) represents one of the most powerful methods to map in vivo protein-RNA interactions. However, the traditional CLIP protocol is technically challenging, which requires radioactive labeling and suffers greatly from material loss during PAGE-Western purifications. Here we introduce a super-efficient CLIP method (GoldCLIP) that omits all gel purification steps. This nonisotopic method allowed us to perform highly reproducible CLIP experiments with multiple RBPs including Ago2 and PTB in human cell lines. In principle, our method guarantees successful library construction as long as the protein of interest can be crosslinked to RNAs in living cells. GoldCLIP is readily applicable to diverse systems and should greatly accelerate RNA related investigations.
