Malonyl-CoA: acyl carrier protein transacylase (MCAT) is a critical enzyme responsible for the transfer of the malonyl moiety to holo-acyl carrier protein (ACP) forming the malonyl-ACP intermediates in the initiation step of type II fatty acid synthesis (FAS II) in cyanobacteria. MCAT has been considered as an attractive drug target in the discovery of antibacterial agents. In this study, 1.8? High Resolution Crystal Structure of Malonyl CoA-Acyle Carrier Protein Transacylase (MCAT) from Synecocystis sp. PCC 6803 is reported. This is the first crystal structure discovered in the family of cyanobacteria and solved in DICP. The processes how to solve the crystal structure and the structural comparison with MCATs from other organism will also be presented. We believe this structure information will greatly facilitate our understanding of the basic mechanism of fatty acid synthesis.

Dr. Yanming Zhang is a Vice President of WaterStone Pharmaceutics and an Invited Foreign Expert of 1812 group in DICP. He has over 25 years of experience in fundamental and applied research on macro molecular X-ray crystallography and structural Biology in topics ranging from protein purification, protein or protein complex crystallization, MR/MAD/SIR/MIR protein structure solution, structural based drug design, to structure/function relation. He has international working experiences in research units and Universities in China, Canada, USA, and Korea. Lately, he has academically cooperated with Prof. Shi Yigong in Tsinghua University and Professor Deng zhixin in Wuhan University. Dr. Zhang has published fourteen papers, 2 U.S. patents, been a core member for the development of 2 new FDA approved drugs, and successfully completed at least 10 research projects.
